MONROVIA, CA, July 09, 2012 (Press-News.org) Marcus Brewster, Vice President of Pharmaceutical Development & Manufacturing Sciences at Janssen R&D (a Johnson & Johnson company) will give the Keynote Presentation on ¡°Improving Oral Bioavailability for Poorly Water-soluble Drug Candidates¡± at the Drug Delivery Technologies & Formulation Conference on Sep 5-6, 2012 in Zurich, Switzerland.
Contemporary pipelines are often highly populated with difficult to formulate drug candidates. The genesis of this complexity is thought to be rooted in drug selection paradigms including HTS screening, the nature of current drug target space and issues associated with drug candidate physical form. Given the challenges, novel formulation technology is evolving to expand appropriate formulation space such that candidates can be presented to the body with a highly oral bioavailability as possible. Technologies that increase dissolution rate include particle size reduction methodologies including nanonization wherein the drug is reduced to sizes in the 100-400 nm range. Solubility enhance technologies include the use of complexing agents such as cyclodextrins and dispersion associated with either amorphous glassy systems or lipid dispersions including self-emulsifying approaches. In all of these cases, technologies to screen for the most appropriate formulation components and processing strategies are required and include the use of miniaturization and automated down-scaled techniques often based on 96-well plate systems. Multiplexed nanomills, high throughput supersaturation screening and solid dispersion assessments, cyclodextrin formulation will find workflows and systems to assess drug lipid and surfactant solubility and emulsification.
Dr. Brewster¡¯s presentation will discuss the current and emerging formulation technical horizon, define formulation space to help target the best enabling technology to a particular formulation need, assess technology approaches for early drug candidates, and assess resource needs as a function of formulation difficulty.
GTC¡¯s Drug Delivery Technologies and Formulation Conference will provide education, communication and networking opportunities for both the scientific and business leaders engaged in drug delivery and formulation. Leaders from academia, biotech and industry will discuss the current challenges and new strategies to succeed in this evolving area.
